    Is it possible to use DisplayCAL  to make ‘spot’ measurements? I would like to manually record the CIE x,y, and the luminance values of computer-generated color patches.

    If you use the testchart editor to enter all the RGB combinations you want to measure, you can do it in one go. The result will be CIE XYZ.

    Thank you Florian! One last question….What if I would like to measure stimuli that are not computer generated (e.g. Pantone cards, color plates)?

    You need an instrument capable of reflective readings for that (typically a spectrometer) and use ArgyllCMS spotread utility on the command line (see ArgyllCMS documentation).
