    Not sure whether I should use Windows’ built-in calibration process for Gamma and Contrast, and if so at which stage of calibration?

    I have a Spyder5Express and would be following this guide using DisplayCal.

    Don’t use Windows calibration process at all. It’s close to trash. It can only correct display gamma in some cases, but it can’t build correct ICC profile. You need to choose appropriate correction for your Spyder, all other things are well done with DisplayCAL, probably except of white point that can be groped by xy coordinates (instead of color temperature). If standard correction set and users’s online base do not contain correction for your display (or similar display), than you need to rent somewhere i1 Pro / Pro 2 or ColorMunki to make correction for your Spyder, DisplayCAL has this function.
