Yes, you can do that. Although note that the goal is usually not to aim for an arbitrary monitor luminance, but to control your environmental lighting and then adjust the monitor so it is neither too dim nor too bright for the environment. Then, after you have established a brightness setting, use “As measured” in DisplayCAL. In a typical home/office environment (during the day), monitor luminance that allows comfortable work usually falls roughly in the 120-160 cd/m2 range.
